If it's a PE file, you can use something like Shellter (you also have the choice to add meterpreter to it).
If it's a .NET file, find a good crypter (best to program one yourself, or buy a top quality one).
Don't upload to virus scanning websites where it will share the detections (VirusTotal for example).
NoDistribution/AntiScan is a good one.

You should also try to avoid suspicious actions such as known UAC bypasses to prevent signature detection as crypters won't always make you FUD.
